小鼠肺细菌感染模型肠道菌群变化的实验研究Experimental study on change of intestinal flora in mice models with pulmonary bacterial infection

摘要(Abstract):
目的 构建小鼠肺细菌(肺炎克雷伯菌、金黄色葡萄球菌、鲍氏不动杆菌)感染模型,分析小鼠肠道菌群的变化。方法 C57BL/6小鼠经气管插管分别诱导感染肺炎克雷伯菌(Kp)、鲍氏不动杆菌(Ab)及金黄色葡萄球菌(Sa),观察肺部炎症,取肺组织匀浆培养进行细菌计数,采用16S rDNA测序技术对粪便菌群进行检测并进行生物学信息分析。结果 感染后第7天,显微镜下可见各感染组小鼠肺部支气管周围均可见大量炎症细胞浸润,肺组织病理评分>3分,肺匀浆病原菌计数均超过105 CFU/g,符合急性肺部感染指征。感染后第3、7天,Kp组、Sa组较PBS组(磷酸缓冲盐溶液对照组)小鼠肠道菌群物种数增加(P<0.05);随感染时间延长,各组间肠道菌群群落结构差异显著增大(P<0.05);随感染时间延长,各组间肠道菌群门水平差异显著增大(P<0.05);各感染组较PBS组小鼠肠道益生菌联合乳杆菌属比例显著降低,NK4A136组、另枝菌属比例显著增加(P<0.05); Kp组较PBS组小鼠肠道益生菌乳杆菌科比例降低(P<0.05)。结论 不同细菌所致肺感染对小鼠肠道菌群群落结构以及物种数有显著影响,不同细菌致肺感染后均导致特定肠道益生菌丰度降低。
